No. 16 at Port Royal in Bermuda from the back tees is by far the scariest/ most intimidating I've ever been on.
#18 on the course where you are about to break 100, 90, 80, 70 or par!
I have never played any of these holes and I am not familiar with #5 to #10 except for Kiawah's 17th. Based on my game, #16 at Cypress would probably be the most intimidating. That's a lot of wind, water and rocks to negotiate.
Brian, your answer is just perfect! any hole when you're about to achieve your goal after ton of work and frustration becomes the hardest due to the pressure...
Also, Stonewater Golf Club #17: a 247yds par 3 from with water in front of the green, OB over the green (not that i can reach it) and bunkers all around... by far the hardest and most intimidating hole i've ever played...

I've played #1 and #2 twice (replicas on Tour 18 in Houston) I birdied #2 once almost holing out by mere inches and and parred the other time and on hole # 1 I parred both. looking back the island is more intimidating then Augusta #12!
I played #17 at TPC Sawgrass twice. Made 2 pars. The shaot was right at 138 yards both times.
